| 摘 要： | 目的建立罗通定血浓度反相高效液相色谱测定方法。方法血浆样品经乙酸乙酯提取,AichromBond-1 C18色谱柱分离;以甲醇-水(75/25,V/V)为流动相,流速1.0ml.min-1,检测波长211nm,进样量30μl。结果罗通定在0.01μg~1μg.ml-1范围内线性关系良好,最低检测限为0.01μg.ml-1,相对回收率为102.2~105.0%,日内RSD≤5.00%,日间RSD≤4.22%。结论该方法简便,准确,灵敏,可用于罗通定血浓度测定。

| Abstract: | OBJECTIVE To establish an RP-HPLC method for determination of rotundine in human plasma.METHODS The plasma samples were extracted by ethyl acetate.The Aichrombond-1 C_(18) column and mobile phase of methanol and water with a ratio of 75∶25 at flow rate of 1.0ml·min~(-1) and the detection wavelength of 211nm were used for rotundine analysis.The injection volume was 30μl.RESULTS The linear range of rotundine was 0.01~1μg·ml~(-1) and the detection limit was 1μg·ml~(-1).The relative recovery was from 100.2 % to 105.0%.The intra-day RSD and inter-day RSD were less than 5.00%and 4.22%,respectively.CONCLUSION The method was simple,accurate,rapid and sensitive.It can be used to determine the rotundine concentration in human plasma. |
