大鼠心肌缺血再灌注损伤中TNF基因表达状况

摘    要:目的检测在心肌缺血再灌注损伤中TNF的表达,并初步探讨其机制。方法采用Langendorff模型及大鼠离体心脏灌注模型,心肌组织缺血30min后、再灌注30、60、120min时,用原位杂交、免疫组化和酶联免疫法检测心肌细胞中TNFrnRNA、蛋白的表达。结果缺血再灌注实验组心肌组织中的TNF蛋白水平比对照组明显升高;实验组心肌细胞中的TNFmRNA和蛋白表达明显,染色呈强阳性或极强阳性(t=2.689,P〈0.05),并且呈阳性表达的细胞是心肌细胞。结论缺血再灌注可刺激心肌细胞大量表达TNF,并且心肌组织中的TNF主要由心肌细胞表达产生。
