自含式压力蒸汽灭菌生物指示剂孢子计数方法的考察

摘    要:摘 要 目的: 研究自含式压力蒸汽灭菌生物指示剂孢子计数方法。方法: 采用活菌计数法,将不同厂家产品在不同洗脱液、不同平皿计数方式下的回收菌数进行计数,并利用单因素方差分析方法(ANOVA)对不同处理组回收菌数的几何均数进行统计学检验。结果: 洗脱液为灭菌纯化水,培养基为胰酪胨大豆琼脂培养基,纸片破碎形式为涡旋振荡法(加入玻璃珠),平皿计数形式采用浇碟法的操作方法能获得较好的实验结果。结论: 在保证自含式压力蒸汽灭菌生物指示剂的孢子计数菌数回收率的情况下采用上述操作能降低实验成本且易于操作。

Abstract:ABSTRACT Objective: To study the methods for spore counting using self contained pressure biological indicator for steam sterilization. Methods: A live bacteria counting method was applied in the recycling bacteria counting using the products from different manufacturers with various eluent and plant counting methods, the ANOVA analysis was used to statistically detect the difference in the geometric average value of the recycling counting among the different treatment groups. Results: Sterilized purified water as the eluent, soybean casein digest agar medium as the culture medium, vortex oscillation (with glass beads) as the broken form and pouring plate operation as the plate counting form would obtain better experimental results. Conclusion:Under the condition described above, the cost is reduced with easy operation and promising recovery of spore counting using self contained pressure biological indicator for steam sterilization.
