白花蛇舌草多糖的提取纯化工艺研究

目的优选白花蛇舌草多糖的提取及纯化工艺。方法采用正交试验设计法优选白花蛇舌草多糖的最佳提取工艺;通过乙醇沉淀法、鞣酸沉淀法并联用活性炭对传统多糖的纯化工艺进行改进。结果最佳提取工艺为:白花蛇舌草药材预先加12倍量水浸泡2h,加热至70℃,提取3次,每次1.5h,采用鞣酸沉淀法联合活性炭可有效去除多糖中的蛋白质和色素等杂质。结论提取时间对多糖的提取效率影响较大;所优选的纯化工艺方法简便,蛋白去除率高,色素脱除完全,多糖损失少,工艺可行性强。

Study on the preparation and purification technology of polysaccharides in Oldenlandia diffusa Willd

OBJECTIVE To optimize the extraction and purification method of the polysaccharides in Oldenlandia diffusa Willd.METHODS The bset extract method of the polysaccharides was determined by orthogonal design.Through the study on the ethanol extraction and tannic acid extraction,the best purification method was chosen.RESULTS The best extraction method of the polysaccharides in Oldenlandia diffusa Willd.was：the immersion time was 2h,the solvent was 12 times water,which heated to 70℃,extracted 3 times and 1.5h each time.Tannic acid extraction with active carbon was used to remove the protein and colouring matter from the polysaccharides.CONCLUSION The extraction time greatly influenced the extraction of polysaccharides.Tannic acid extraction was used to make the process simple and the high removal rate of protein.The extraction and purification technology of Oldenlandia diffusa Willd.is feasible.
