温育时间对酶联免疫吸附试验结果的影响

目的 探讨温育时间对FAME全自动酶免分析系统(以下简称FAME)进行酶联免疫吸附试验(ELISA)结果的影响,从而提高ELISA法检测结果的准确性.方法 根据FAME孵育巢温度变化曲线,延长ELISA温育时间,选择不同实验参数条件进行对照实验,再根据实验结果选择合适的补偿时间进行实验,所得数据采用SPSS 13.0 for Windows进行双变量相关分析.结果 乙型肝炎表面抗原阳性标本、阳性对照、0.5 ng/ml质控对照和1 ng/ml质控对照的吸光度(OD)值与温育时间延长呈正相关,阴性标本和阴性对照的OD值与温育时间延长无统计学意义.结论 适当延长ELISA温育时间,可提高FAME结果的准确性.

Effect of incubative time for the accuracy of ELISA

Objective To explore the effect of incubative time to ELISA assay with FAME system,and to im- prove the accuracy of ELISA assay results.Methods The curve of temperature change in FAME incubator slot was traced.Different test parameters were set according to the temperature change curve for the control experiment,and then corresponding incutative time was chosen based on the ELISA assay results.Bivariate correlation analysis about the test data was performed with SPSS 13.0 software for Windows.Results The optical density value of HBsAg positive samples,positive control samples,1 ng/ml of quality control samples and 0.5 ng/ml of quality control sam- ples were positive correlative to the incubative time.The optical density value of negative samples and negative con- trol samples were not correlative to the incubative time.Conclusion The accuracy of ELISA test with FAME system might be improved by properly prolonging the incubative time.
