摘    要:目的 观察5-烯丙基-7-二氟亚甲基白杨素(ADFMChR)体外诱导人宫颈癌HeLa细胞凋亡的作用效果及其可能机制.方法 体外培养人宫颈癌HeLa细胞,分别应用不同浓度的ADFMChR处理人宫颈癌HeLa细胞,用AO/EB染色观察ADFMChR作用HeLa细胞48h后凋亡形态学改变;流式细胞术(FCM)检测ADFMChR作用HeLa细胞后的凋亡率;Westernblot分析ADFMChR对HeLa细胞caspase-3蛋白和mcl-1蛋白表达的影响.结果 不同浓度的ADFMChR处理HeLa细胞48h后,细胞出现典型的细胞凋亡形态特征,凋亡率呈浓度依赖性,同时细胞caspase-3蛋白表达上调,mcl-1蛋白表达下降.结论 ADFMChR具有体外诱导人宫颈癌HeLa细胞凋亡作用,呈浓度依赖性,其机制可能与其上调caspase-3及下调mcl-1蛋白表达有关.

Abstract:Objective To investigate the effects of 5 - Allyl - 7 - Gem - Difluoromethoxy - chrysin(ADFMChR) and possible mechanism on apoptosis of human cervical cancer HeLa cells cultured in vitro. Methods Apoptosis morphologic changes of HeLa cells were observed by AO/EB double fluorescence stain. The apoptosis rate of HeLa cells was analyzed by FCM using PI staining. The expressions of caspase - 3 and mcl - 1 proteins were analyzed by western blot. Results Typical morphologic changes of apoptosis in HeLa cells could be observed after treatment with different concentrations of ADFMChR by fluorescence microscope using AO/EB fluorescence staining. FCM showed that the apoptosis of HeLa cells was induced by ADFMChR in a concentration - dependent manner Western Blot a- nalysis showed that ADFMChR downregulated expression of Mcl - 1 protein and upregulated expression of caspase -3 protein. Conclusion ADFMChR could effectively induce apoptosis of HeLa cells cultured in vitro, in a concen- tration- dependent manner,which maybe associate with up -regulating caspase -3 expression and down -regula- ting mcl - 1 expression.
