姜茎尖的离体培养与试管苗快繁

摘    要:目的 探索姜茎尖组织培养及其试管苗快速繁殖的适宜条件。方法 剥取不大于1mm的姜茎尖生长点为外植体,以MS为基本培养基,附加不同种类和浓度的植物生长物质进行培养。结果 小于0.3mm的姜茎尖可进行良好的分化与生长;在各类激素中,以BA对姜芽分化最为有效,培养基MS BA 1.0mg/L NAA 0.2~0.4mg/L最有利于芽的分化和生长;在生根培养基中添加适量的BA不仅不影响不定根的形成,且能增加成苗数,对不定根的分化和成苗最适的培养基为1/2 MS NAA 0.1mg/L BA 0.15mg/L。结论 试验为姜的茎尖离体培养提供了有效途径。

Abstract:Object To study the suitable conditions on tissue culture of stem apex and shoot rapid propagation of ginger. Methods The ginger stem apex ((?)<1 mm) was taken as explants and cultured on MS medium with different portions of plant growth substances. Results The ginger stem apex thinner than 0. 3 mm in diamond could differentiate and grow very well. The optimal medium for shoot propagation and growth was MS BA 1. 0 mg/L NAA 0. 2-0. 4 mg/L. Adding suitable amount of BA to root medium did not influence the formation of adventitious root, but also increased the seedling formation, and the suitable medium for adventitious root induction and seedling formation was 1/2 MS NAA 0. 1 mg/L BA 0. 15 mg/L. Conclusion It is to provide the effective ways to culture stem apex of ginger in vitro.
