Caspase-9和Bax在胃癌中的表达以及与凋亡的关系

摘    要:目的:探讨Caspase-9和Bax在胃癌发展中的作用以及二者的表达与细胞凋亡的关系。方法:应用免疫组织化学S-P法检测Caspase-9和Bax在57例胃癌及48例非癌胃黏膜组织的表达,用原位末端标记法(TUNEL法)检测相应胃组织细胞凋亡。结果:Caspase-9蛋白在胃癌组织中阳性表达率为40.35%,较非癌胃黏膜组织的阳性表达率70.83%明显降低(P〈0.05);Bax蛋白在胃癌组织中阳性表达率为35.09%,明显低于非癌胃黏膜组织的72.92%(P〈0.05)。Caspase-9蛋白阳性表达者细胞凋亡指数(AI)值(11.20±3.39)%与其阴性者AI值(6.22±2.45)%比较差异有显著性(P〈0.05);Bax蛋白阳性表达者AI值(12.54±3.60)%,与其阴性者AI值(7.18±1.25)%比较差异有显著性(P〈0.05)。在不同类型胃癌组织中,高中分化腺癌、低分化腺癌、黏液癌的AI值分别为(5.72±1.25)%、(2.54±0.98)%、(3.28±1.42)%,其中高中分化腺癌和后二者之间有统计学差异(P〈0.05)。结论:Caspase-9和Bax可能与胃癌的发生或发展相关;推测Bax激活Caspase-9的线粒体通道受抑可能参与胃癌的发生。

Abstract:Objective: To explore the effects of caspase-9 and bax in the development ot stomach cancer and the relation of their expression to cell apoptosis. Methods: Expression of caspase-9 and bax in 57 specimens of stomach cancer and 48 specimens of non-cancer stomach tissue was detected with immunohistochemistry S-P method, and the cell apoptosis in stomach tissue detected with TUNEL method. Results: The positive expression rate of caspase-9 in stomach canser tissue (40.35%) was significantly lower (P 〈 0.05) than that in non-cancer stomach tissue (70.83%). The positive expression rate of bas in stomach canser tissue (35.09%) was significantly lower (P 〈 0.05) than that in non-cancer stomach tissue (72.92%). The apoptosis index (AI) of easpase-9 positive eases ( 11.20 ± 3.39 ) % was different from that of caspase-9 negative cases (6.22 ± 2.45 ) %, and the difference was significant (P 〈 0.05 ). The AI of bax positive cases (12.54± 3.60)% was different from that of bax negative cases (7.18 ± 1.25 ) %, and the difference was significant (P 〈 0.05 ). AI values in well-differentiated adenocarcinoma (WDA), poorly differentiated adenocarcinoma (PDA), and colloid carcinoma ( CC ) were ( 5.72±1.25 ) %, (2.54 ± 0.98 ) %, and ( 3.28 ± 1.42) % respectively. There were significant differences between WDA AI and PDA AI, and between WDA AI and CC AI (P 〈0. 05). Conclusions: Caspase-9 and bax might be related to the occurrence and development of stomach cancer. The inhibition of mitochondrial channel for caspase-9 stimuted by bax might participate the initiation of stomach cancer.
