女贞子对自由基损伤的红细胞膜流变性的保护作用——分子学机理探讨

摘    要:目的:为探明女贞子对自由基损伤红细胞膜流变性的保护作用的分子机理。方法:对自由基作用前后红细胞膜分子流动性和分子结构的改变与自由基加女贞子提取液共同作用后的红细胞膜相应参数进行了比较。结果:加入女贞子提取液后:(1)自由基产生率平均降低了64%以上。(2)红细胞膜剪切弹性膜量只增加27.7%。(3)对自由基引起的膜蛋白分子和膜脂分子流动性降低的保护不甚明显。(4)能够防止自由基对氢键的破坏,但不能防止二硫键形成所造成的膜蛋白结构的重构。(5)不能减轻自由基对膜磷脂亲水头部和疏水尾部的过氧化作用。结论:本研究中自由基损伤程度与自由基浓度有一定的量效关系,并在膜脂分子损伤后3h 表现出部分可逆性,但与自由基浓度相关。女贞子可减轻自由基对膜分子的损伤程度。

Abstract:Objective:To investigate the molecular mechanism of Fructus Ligustri Lucidi (RLL) in protecting membrane rheo- logical characteristics of red blood cell from free radical damage.Methods:The changes in membrane molecular chemical structure and their fluidity after free radical attacking for 30 minutes were observed and compared to that of protected group by RLL.Results:In the RLL group:(1) The productive rate of free radical lowered for at least 64% in average; (2) The membrane elastic shear modulus increased for 27.7% only;(3) Protective effect on either membrane protein or lipid molecular fluidity was not significant;(4) RLL could prevent damage of hydrogen bond from free radical attacking but couldn't prevent membrane protein reconstitution from disulphic bond formation.(5) Peroxidation in either mem- brane hydrophobic or aqueous areas was not limited.Conclusion:Degree of membrane damage was related to the concen- tration of free radical,it could be partly restored after 3 hrs attacking,the recovery was dependent on free radical concen- tration.RLL could relieve the damage on cell membrane to a certain degree.
