大肠癌组织中ERCC1、TS表达及临床意义

摘    要:目的:探讨ERCC1和TS蛋白在大肠癌中的表达及其临床意义。方法:采用免疫组化SP法和组织芯片对196例大肠癌组织和50例正常大肠切缘组织进行ERCC1和TS蛋白表达水平检测,分析两者与临床病理特征的关系。其中129例术后患者行FOLFOX4方案化疗,随访3年。结果:在结直肠癌中ERCC1及TS表达均高于正常肠黏膜组织,而与性别、年龄、分化程度、部位无关(P〉0.05),TS阳性表达率及两者双阳性表达率均在有淋巴结转移者及TNMⅢ期、Ⅳ期患者中明显升高。ERCC1、TS阴性表达者3年DFS均分别高于阳性表达者(P〈0.05)。结论:ERCC1和TS蛋白在大肠癌中的阳性表达较高,可能与肿瘤的发生、转移有关。ERCC1和TS蛋白阴性者可从FOLFOX4化疗中获益。

The Expression of TS,ERCC1 in Colorectal Carcinoma and Their Clinical Significance

Abstract:Objective:To evaluate the expression of ERCC1 and TS protein in colorectal carcinoma and its clinical significance.Method:TS protein in 196 cases of colorectal cancer tissues and 50 cases of normal colorectal tissue were applied SP immunohistochemistry and tissue microarray to detect the expression level of ERCC1.The relationship between both of them and the clinical pathological characteristics were analyzed.129 cases of patients were treated with FOLFOX4 chemotherapy and were followed up for 3 years.Result:In colorectal cancer the expression of ERCC1 and TS were higher than those of normal intestinal mucosa,regardless of gender,age,degree of differentiation and part( P 0.05).The positive expression rates of TS and two double positive expression rate in lymph node metastasis and TNM stage Ⅲ,Ⅳ patients increased significantly.The 3 years DFS in negative expression of ERCC1,TS were higher than of the patients of positive expression respectively( P 0.05).Conclusion:The expression of ERCC1,TS protein in colorectal carcinoma are higher than normal tissue.It may be associated with tumorigenesis and tumor metastasis.The patients of ERCC1,TS protein negative expressed could benefit from FOLFOX4 chemotherapy.
