胎儿动脉平滑肌细胞培养方法的研究

摘    要:目的:对胎儿动脉平滑肌细胞(hASMC) 培养方法进行研究。方法:贴块法、酶解法、贴块+ 酶解法。结果:3 种方法成功培养了胎儿动脉平滑肌细胞,培养的细胞呈典型“峰 谷”样生长,透射电镜下可见胞浆内含有肌丝束及其相联的致密体。结论:贴块法培养hASMC生长周期较长,但成功率较高;酶解法的消化程度不易掌握,但其生长周期较前者短4~5 d;二者相结合法的贴块培养细胞从组织块中迁移萌出时间比常规贴块法短1 ~2 d。

Abstract:Objective:To study on the techniques of cultured fetus arterial smooth muscle cells(hASMCs).Methods:Explant culture,enzyme ispersed culture and combined explant culture with enzyme dispersed culture.Results:The primary culture of hASMC was performed successfully by using the three methods mentioned above.The cells growed into “hill valleys” characteristicly;the bundle of myofilaments and densities was observed in the cytoplasm of culture the hASMC under an electron microscope.Conclusions:The explant cultured hASMC easily grew,and growth period of the cells was longer than enzyme dispersed SMC in culture (4~5 d),but the degree of enzyme dispersion wasnt easily grasped.The cells of the third culture method migrated from explant earlier than the cells of routine explant culture method.
