槲皮素对小鼠胶原诱导性关节炎症的影响

摘    要:目的探讨槲皮素对Ⅱ型胶原诱导性关节炎(CIA)小鼠关节炎症的影响。方法 6-7周龄的DBA/1 J小鼠随机分为4组,每组10只,对照组,模型组,甲氨喋呤组,槲皮素组。除对照组外,其余小鼠给予牛属Ⅱ型胶原+完全弗氏佐剂进行CIA小鼠第一次免疫,第21天给予牛属Ⅱ型胶原+不完全弗氏佐剂进行加强免疫,次日各组开始给药30天,观察小鼠体重、关节炎指数、足掌厚度、关节病理变化指标。结果与模型组相比,槲皮素可显著改善CIA小鼠关节炎指数、足掌肿胀,对CIA小鼠体重无明显影响,可显著改善CIA小鼠踝关节的病理表现,减轻踝关节的骨破坏。结论槲皮素可有效的减轻CIA小鼠关节滑膜炎症,减轻关节骨破坏,槲皮素是治疗类风湿性关节炎的潜在有效药物。

Abstract:Objective To investigate the effect of quercetin on joint inflammation in type II collagen-induced arthritis (CIA) in mice. Methods DBA/1J mice aged 6-7 weeks were randomly divided into 4 groups, control group, model group, methotrexate group, and quercetin group, with 10 mice in each group. Except the mice in the control group, mice in other groups received the first immunization of bovine type II collagen and complete Freund''s adjuvant. The bovine type II collagen and incomplete Freund''s adjuvant was boosted on the 21st day. The mice in each group received corresponding drugs from the next day and lasted for 30 days. The body weight, arthritis index, paw thickness, and pathological changes of the joint were observed. Results Compared with the model group, quercetin significantly relieved the arthritis index and paw swelling of CIA mice, but had no significant effect on body weight of CIA mice. It significantly relieved the pathological performance of ankle joints in CIA mice and reduced the bone destruction of the ankle joints. Conclusion Quercetin effectively reduces synovial inflammation in CIA mice and reduces bone destruction. Quercetin is a potential drug for rheumatoid arthritis.
