黄芩苷对H_2O_2诱导的SH-SY5Y细胞损伤的保护作用及其对Trx表达的影响

摘    要:目的探讨黄芩苷对H2O2诱导的SH-SY5Y细胞损伤的保护作用及其作用机制。方法采用体外培养细胞的方法,建立SH-SY5Y细胞H2O2损伤模型。分设药物组(50,100,200μmol/L)、H2O2损伤组、阴性对照组。采用MTT法检测细胞活力,采用实时荧光定量PCR法检测各组细胞的Trx表达的变化。结果同H2O2损伤组比较,黄芩苷能明显减轻H2O2引起的SH-SY5Y细胞的损伤,同时Trx表达升高。结论黄芩苷对H2O2诱导的SH-SY5Y细胞损伤具有明显的保护作用,其作用机制可能是黄芩苷通过上调Trx表达,起到抗氧化作用,继而起到抗凋亡的作用。

Abstract:Purpose To study the effect of baicalin on H2O2-induced SH-SY5Y cell injury and its mechanism.Methods Using SH-SY5Y cell line to establish H2O2-induced SH-SY5Y cell injury model.We design four groups(0,50,100,200 μmol/L) and test cell viability by MTT assay and Trx mRNA expression levels by Real-time quantitative PCR.Results Compared with the H2O2 injury group,baicalin can significantly reduce the H2O2-induced SH-SY5Y cell damage,while Trx,expression was increased.Conclusion Baicalin can protect SH-SY5Y cell from H2O2-injury and its mechanism of baicalin-protected may be increased Trx,and then play the anti-apoptotic role.
