侧柏叶炮制方法研究

摘    要:目的:研究侧柏叶炮制前后槲皮素含量的变化,建立侧柏叶合理的炮制方法。方法:通过HPLC法对侧柏叶中槲皮素的含量进行测定,色谱柱为Agilent Eclipse XPB-C18(4.6 mm×250 mm,5μm);流动相为甲醇:0.2%磷酸溶液(55∶45);流速1.0 mL·min^-1;检测波长360 nm;柱温30℃;并对侧柏叶的生品、烘制品及不同炮制品中槲皮素进行测定和比较。结果:槲皮素在0.03~0.52μg成良好的线性关系,回收率为97.5%~100.8%,RSD1.22%,槲皮素在烘制品中的含量较传统炮制品高。结论:所建立的HPLC法简便可靠、精密度高。烘制法具有和传统炮制法相接近的结果,提示其作为侧柏叶的炮制法存在一定的可行性。

Study of Cacumen Platycladi Leaf Processing Method

Abstract:Objective:To study the variation in quercetin content before and after Cacumen Platycladi processing and to establish reasonable processing method.Methods:By HPLC method,we determined the content of quercetin in Cacumen Platycladi,chromatographic column of Agilent Eclipse XPB-C18(4.6 mm×250 mm,5μm);the mobile phase of methanol:0.2% phosphoric acid solution(55∶45);the flow rate was 1.0 mL·min-1;the detection wavelength was 360 nm;column temperature was 30℃;and the effect of quercetin on raw Cacumen Platycladi,baking products and its different traditional processed products were determined and compared.Results:Quercetin in a good linear relationship between 0.03~0.52 μL,the recovery rate was 97.5%~100.8%,RSD=1.22%.The content of quercetin in baked products was higher than that in traditional processed products.Conclusion:The established HPLC method is simple and reliable with high precision.The baking method has similar results with traditional processing method.It is feasible as a processing method on Cacumen Platycladi.
