摘    要:目的 探讨重组人血小板生成素 (rhTPO)体内的生物学活性。方法 本研究以Babl/c纯系小鼠为实验对象 ,将其分成实验组及对照组 ,实验前采尾静脉血 ,测小鼠血小板正常值 ;然后连续 7d腹腔注射rhTPO ,并于注射后第 7天及第 14天采眶静脉血 0 .2ml,测定小鼠外周血小板数量 ,同时观察小鼠的死亡情况。结果 在实验第 7天 ,实验组小鼠外周血小板数明显高于同期对照组 (P <0 .0 5 ) ;而且随实验时间的延长 ,实验组血小板数有升高趋势。结论 rhTPO促进小鼠外周血小板的产生 ,增加外周血小板数量。

Abstract:Objective To investigate the biolgical activity of recombinant(rhTPO) human thrombopoietin in vivo.Methods Normal peripheral platelet counts were performed before experiment on sample obtained from the tail vein of purebred Babl/mice diveded into experiment group and control group.rhTPO was administered intraperitoneally for 7 days.On the seventh and the fourteeth day after injection platelet counts were performed on 0.2ml sample obtained from the supra-obitalis vein.Results On the seventh day and the fourteenth day,the platelet counts of mice treated by rhTPO were higher than by PBS(p<0.05).Moreover the platelet counts of mice in experimental group of rhTPO showed increasing direction following experimental days.Conclusions TPO has obvious biological activity in increasing platelet production.
