肿瘤转移抑制基因nm23H_1产物在人鼻咽癌中表达的研究

摘    要:采用SP免疫组化技术,以NDPK/nm23H1特异的单克隆抗体为探针,对人鼻咽癌中nm23H1的表达及其与颈淋巴结转移的关系进行了研究。结果:31例鼻咽癌中nm23H1基因产物表达的阳性率为41.9%(13/31)。其中无转移组的阳性率为52.3%(11/21),有转移组的阳性率为20%(2/10),两组间具有显著性差异(P<0.01),而在转移的颈淋巴结组织中nm23H1产物表达极微或无表达,其阳性率为0,与无转移组比较有显著性差异(P<0.01)。上述结果说明,肿瘤转移抑制基因nm23H1产物的表达,在无转移组呈高表达,在有转移组及淋巴结转移组中呈低表达,其表达与转移呈负相关,说明人鼻咽癌的转移与nm23H1基因的低表达密切相关

Expression of Metastasis Suppressor Gene nm23H 1 Product in Nasopharyngeal Carcinoma

Abstract:Using the specific monoclonal antibody of nm23H 1 gene product and the immunohistochemical technique, we studied the expression of DNPK/nm23H 1 and its correlation with lymph node metastasis in nasopharyngeal carchnoma (NPC). The results revealed that, in a total of 31 cases of NPC, 13 showed positive expression with a positive rate of 41.9%. Among 21 cases of NPC with no lymph node metastasis, 11(52.3%) showed positive expression while only 20% (2/10) of those with lymph node metastasis were positive, and there was significant difference between them ( P <0.01). Meantime the expression was very low or not detectable in the cancerous cells of the match lymph node metastasis and all 10 cases showed negative expression. Statistical significant difference was found between the group of NPC with no metastasis and the group of lymph node metastasis ( P < 0.01). The above results indicated that high expression of DNPK/nm23H 1 existed in the group of NPC with no metastasis while low expression existed in those with metastasis and lymph node metastasis. It suggests that the expression of nm23H 1 is inversely associated with lymph node metastasis. The DNPK/nm23H 1 may play a role in polymerization of microtubulin protein and participate in the process of cancer metastasis.
