p185~(neu)和p21~(ras)在肺癌组织中表达及相互关系

摘    要:目的 检测 c- erb B- 2基因产物 p185 neu和 ras基因产物 p2 1ras在肺癌组织的表达情况 ,以揭示 c- erb B- 2和 ras基因在肺癌形成中的作用及其相互关系 .方法 运用免疫组织化学 SABC法检测 5 5例原发性肺癌组织中 2种基因产物 .结果  10例小细胞肺癌组织均无 p185 neu和 p2 1ras的阳性表达 .2 8例肺鳞癌中 p185 neu和 p2 1ras阳性表达率分别是 46 %(13/ 2 8)和 5 4% (15 / 2 8) .16例肺腺癌中 p185 neu阳性表达率为 5 6 % (9/ 16 ) ,p2 1ras阳性表达率为 6 2 % (10 / 16 ) .肺鳞癌和肺腺癌中 2种蛋白的阳性表达率均无显著性差异 (P>0 .0 5 ) .p185 neu阳性表达与淋巴结转移具有明显相关性 (r=0 .36 ,P<0 .0 5 ) ,与其他临床病理因素无关 ;p2 1ras阳性表达与淋巴结转移、肿瘤大小等临床病理因素无关 .p185 neu和 p2 1ras在肺癌组织中的表达水平呈正相关 (r=0 .5 2 ,P <0 .0 1) .结论 c- erb B- 2和 ras基因的改变与非小细胞肺癌 (NSCL C)的发生有关 ,这 2种基因在 NSCL C的形成和发展中具有协同作用 .同时 ,检测 p185 neu和 p2 1ras还有助于评估肺癌患者的预后

Abstract:AIM To shed light on the relationship among c erb B 2 gene, ras gene, and the carcinogenesis of lung cancer by measuring p185 neu and p21 ras (the products of the c erb B 2 gene and ras gene respectively) in lung cancer tissues.METHODS Two genes' products were detected in primary lung cancer tissues by SABC, an immunohistochemical method, for 55 cases. RESULTS Of 10 cases of small cell lung cancer (SCLC), none was p185 neu or p21 ras positive. And of 28 cases of squamous carcinoma, the rates of p185 neu and p21 ras positive expression were 46% (13/28) and 54% (15/28) respectively. Of 16 cases of adenocarcinoma, the rates of the two genes' positive expression were 56% (9/16) and 62% (10/16) respectively. There was no significant difference between the positive expression rates of the two genes in the squamous and adenocarcinoma ( P >0.05). The p185 neu positive expression was significantly correlated ( r =0.36, P <0.05) to lymph node metastasis but not significantly correlated to other clinical pathological factors. However, p21 ras positive expression had no significant correlation with such clinical pathological factors as lymph node metastasis and the size of tumor. The expression of p185 neu and p21 ras was positively correlated ( r =0.52, P < 0.01 ) in lung cancer tissues. CONCLUSION Change in c erb B 2 gene and ras gene is related to the carcinogenesis of non small cell lung cancer and plays a cooperative role in the carcinogenesis and development of NSCLC. Detecting p185 neu and p21 ras is useful for the evaluation of the prognosis of lung cancer patients.
