Ras相关核蛋白在肺鳞癌组织中的表达及意义

摘    要:目的探讨肺鳞癌组织中Ras相关核蛋白(Ran)的mRNA和蛋白表达水平变化及其意义。方法采用实时荧光定量PCR(RQ-PCR)和Western Blot技术检测22例不同分期及分化程度的肺鳞癌组织及其相应癌旁正常组织中Ran的表达水平,并分析其与临床病理指标的关系。结果肺鳞癌组织中Ran mRNA表达水平是癌旁正常组织的2.35倍(P0.05),Ran在肺鳞癌组织中的蛋白表达水平高于癌旁组织。Ran在非早期(Ⅲ期)肺鳞癌组织中mRNA和蛋白的表达水平均高于早期(Ⅰ期+Ⅱ期)癌组织(P0.05),在低分化肺鳞癌组织和高、中分化癌组织中mRNA和蛋白表达水平的差异无统计学意义(P0.05)。结论 Ran的表达上调可能与肺癌的发生、发展相关。

Abstract:Objective To investigate the expressions on mRNA and protein levels and clinical significance of Ras-related nuclear protein(Ran) in lung squamous carcinoma tissues.Methods Expressional levels of Ran were detected by the methods of real-time quantitative PCR and Western Blot consisting of 22 cases of lung squamous carcinoma tissues and adjacent normal tissues with different TNM stages and differentiated degree,then the correlations of Ran expressional levels to pathological characters were evaluated.Results The mRNA expression of Ran in squamous cell carcinoma tissues was 2.35 times of that in the adjacent normal tissues and the difference was significant(P<0.05).The expressional level of Ran protein in lung squamous carcinoma tissues was higher than that in the adjacent normal tissues.The expressional level on mRNA and protein in Ⅲ stage tissues was higher than that of Ⅰand Ⅱstages in lung squamous cell carcinoma(P<0.05).The difference of expression level between differentiated degree was not found(P>0.05).Conclusion The increased expression of Ran in lung squamous cell carcinoma might be correlated with carcinogenesis and used as a supporting marker for early diagnosis of lung carcinoma.
