淫羊藿苷对大鼠颈总动脉损伤后基质 金属蛋白酶-2表达的影响

摘    要:目的:研究淫羊藿苷对大鼠颈动脉损伤后基质金属蛋白酶(MMP-2)mRNA表达的影响。方法:取大鼠44只,随机分为正常对照组8只,淫羊藿苷高、低剂量组和模型组各12只。后3组建立鼠颈动脉球囊损伤术后狭窄模型,淫羊藿苷组术前6 d开始ig淫羊藿苷60,120 mg.kg-1至实验结束(14 d和28 d);模型对照组以等体积生理盐水ig。术后14 d和28 d分批处死动物。取各组实验动物颈动脉损伤段,常规制片,免疫组织化学S-ABC法检测MMP-2蛋白的表达;RT-PCR检测MMP-2 mRNA的表达。结果:在球囊损伤后14,28 d时,淫羊藿苷干预组的MMP-2 mRNA及蛋白表达量显著低于模型组(P<0.01)。各干预组之间无明显差异。结论:淫羊藿苷在动脉损伤后早期可通过下调MMP-2 mRNA的表达,抑制平滑肌细胞的迁移,从而减轻细胞外基质(ECM)的合成而抑制球囊损伤后新生内膜的增生。

Abstract:Objective: To observe the effect of icariin(ICA) on the levels of matrix metalloprotease\|2(MMP-2) in rat carotid artery injured by balloon and to evaluate the mechanisms of inhibiting intima hyperplasia and vascular remodeling. Method: Fourty-four rats were randomly divided into 4 groups: natural group, model group and ICA low dose group(60 mg·kg-1) and ICA high dose group (120 mg·kg-1). Rat carotid balloon injury restenosis model was established. Rats of ICA group were treated with(ig) drugs for 2 or 4 weeks. Then the mRNA expression of MMP-2 was assessed by RT-PCR, and immunohistochemistry analysis was applied to detect the expression of MMP-2. Result: Compared with model group, the ICA group significantly decreased the expression of MMP-2, at 14 and 28 day after injury (P<0. 01). Conclusion: ICA could inhibit neointimal proliferation of rat carotid artery injured after balloon injury, the mechanism might be related to the effects of ICA in decreasing mRNA level of MMP-2, inhibiting migration of vascular smooth muscle cells.
