摘    要:目的 研究西洋参茎叶总皂甙(PQS)对人视网膜色素上皮细胞(RPE)增生的抑制作用。方法 通过活细胞计数法与MTT比色法分别检测不同质量浓度(10μg/ml~500μg/ml)PQS和PQS(200μg/ml)在不同作用时间(6~120h)对RPE增生及代谢的影响。结果 PQS组细胞增殖受抑制,细胞生存率下降,400μg/ml抑制作用最强;其抑制效应在6h出现,72h达高峰。结论 PQS可能通过阻滞钙通道、干扰RPE代谢对RPE增殖具有剂量和时间依赖方式的抑制作用。

Abstract:Objective To investigate the effects of panax quinquefolium saponins ( PQS) on proliferation of cultured retinal pigment epithelium (RPE) cells. Methods Cultured RPE cells were treated with PQS by various concentrations (10-500 ug/ml) and PQS 200 ug/ml at different times(6-120 hr). Proliferation and metabolism of RPE cells were evaluated by cell count method and MTT colorimetric assay. Results The survival rate of cultured RPE cells lowered in PQS group. The optimal effective concentration of PQS was 400 u,g/ml. The significant inhibitory effect of PQS occurred at 6 h and the strongest inhibition at 72 h. Conclusion PQS can inhibit the proliferation of RPE cells in dose-dependent and time-dependent manner by blocking calcium channel and obstructing the metabolism of RPE cells.
