THE ASSOCIATION BETWEEN THE POLYMORPHISM OF BRCA1 RELATED MICROSATELLITES AND THE BIOLOGICAL BEHAVIORS OF SPORADIC BREAST CANCER

PURPOSE To study the frequency of LOH (loss of heterozygosity) and MSI (microsatellite instability)of fourBRCAl related polymorphic markers in sporadic breast cancer and to correlate their incidence with clinicopathological parameters. METHODS Polymerase chain reaction-polyacrylamide urea gel electrophoresis-DNA silverstaining was used. RESULTS The total incidence of LOH and MSI of four BRCAl related polymorphic markers inbreast cancer were 62. 5 % and 45. 0%,respectively. The incidence of MSI in patients with tumours 2 cm in diameter or less was higher than those with tumours more than 2 cm in diameter (P<0. 05). There is a tendency for theincidence of MSI to be higher in stage I breast cancer patients than later stage cases,while the contrary exists forLOH. The incidence of LOH and MSI was not correlated to menstrual,lymphnode and estrogen receptor status.CONCLUSION Breast cancer susceptibility gene was located near these four microsatellites DNA. MSI may be anearly event in mammary tumorigenesis, while LOH may occur at later stage.
