McAb检测囊虫病人循环抗原的研究

应用ＭｃＡｂ双抗体夹心－ＥＬＩＳＡ检测脑囊虫病人血清和脑脊液（ＣＳＦ）中循环抗原（ＣＡＧ）。血清经ＰＥＧ沉淀浓缩沸浴和ＣＳＦ经沸浴处理后，ＣＡｇ的阳性率分别为９２．７３％和９０．７４％，总阳性率为９２．０７％，灵敏度６８．７ｎｇ／ｍｌ。对照组中除２例包虫病人血清有交叉反应外，其余均为阴性。平行检测病人ＣＳＦ的ＣＡｇ和ＣＡｂ，以及同步检测血清和ＣＳＦ中的ＣＡｇ，表明它们之间有互补和相互验证的作用，可提高阳性率。对囊虫病人治疗前后血清ＣＡｇ的动态检测，表明血清ＣＡｇ的阴转率随疗程的增加而提高。这些结果说明ＣＡｇ检测不仅可用于囊虫病的诊断，而且可作为疗效考核的重要手段。也说明ＭｃＡｂ双抗体夹心ＥＬＩＳＡ检测ＣＡｇ具有敏感性高、特异性强、重复性好等优点。

DETECTION OF CIRCULATING ANTIGENS FROM THE PATIENTS WITH CYSTICERCOSIS BY McAb

In this paper, monoclonal antibody based on double antibody sandwich ELISA (McAb-ELISA)was used for detecting the circulating antigen (CAg) in sera and cerebrospinal fluid (CSF) from patients with cysticercosis. The positive rate, depended on the treatment method of sera or CSF, was 92.73% after sera were precipitated by polyethylene glyco (PEG) then boiled in water bath, and was 90.74% after CSF was boiled in water bath. The total positive rate of sera and CSF were 92.07% (151/164). The sensitive scale was 68.7ng/ ml. In the control group, the tests were all negative except two miner positive reaction in sera from the patients with echinococcosis. It indicated that the specificity of CAg detecting had improved by using McAb-ELISA.Synchronous detecting the CAg and CAb in Patients' sera, or the CAg in both sera and CSF, the results could be verified and complemented each other, then an increased positive rate coule be expected.The dynamic observation of CAg in sera from patients with cysticercosis before and after treatment indicated that the negative-conversion rate of CAg in sera increased with the treatment course accumulated.These results indicated that CAg detection is very useful in the diagnosis of cysticercosis and in the evaluation of drug efficacy.
