摘    要:目的建立神经千细胞分离、培养方法。方法从新生大鼠海马组织分离神经干细胞,进行体外培养、传代,应用免疫荧光细胞化学方法观察鉴定神经干细胞和分化后神经细胞抗原的表达。结果从海马组织中分离出的神经千细胞具有增殖能力,可进行传代培养,获得的细胞克隆表达Nestin阳性,分化后的细胞表达神经元、星形胶质细胞和少突胶质细胞的特异性抗原。绪论分离堵养的细胞具有自我更新、增殖和多分化潜能,是神经干细胞。

Abstract:Objeetive To establish a method to isolate, culture and identify the neural stem cells from postnatal rat hippocampus. Methods The neural stem cells were isolated from postnatal rat hippocampus, cultured in vitro, and passaged successively. Either the proliferation of neural stem cells or differentiation into new neural and glial cell types could be identified using the immunofluorescence. Results The neural stem cells isolated from postnatal rat hippocampus possess the proliferation capacity and can be passaged successively. The cloned cells were nestin-positve and differentiated cells express characteristic antigen of neurons, astrocytes and oligodendrocytes. Conclusion The cells isolated and cultured from postnatal rat hippocampus have shown a strong ability of self-renewal, proliferation and multipotency of differentiation, which are evidently identified as the neural stem cells.
