| 摘 要： | 目的 调查中国汉族肾脏移植人群mdr1基因C3435T单核苷酸多态性与口服FK506后血药浓度的关系.方法 采用不同引物进行PCR扩增了17例中国汉族人群的基因组,判断人群的C3435T基因型,结合其口服FK506 12 h后血药浓度,判断二者是否存在关联.结果 口服相同剂量的FK506以后,基因类型为T/T的患者血药浓度最高,C/T次之,C/C最低,三者存在显著差异(P＜0.05).结论 mdr1基因的C3435T与口服FK506后的血药浓度密切相关,检测汉族肾脏移植患者mdr1基因的C3435T,对指导临床上FK506的使用有一定帮助.

| Abstract: | Objective To investigate the relationship between the mdr1 gene C3435T polymorphism and the blood concentration of tacrolimus(FK506) after oral administration.Methods PCR was used to amplify the target gene from 17 Chinese Han patients after renal transplantation.The FK506's blood concentration was detected 12 h after oral application.Then the relationship between them was estimated.Results As for those patients who had taken FK506 at the same dose,the blood concentration was highest in those of T/T genotypes,and then C/T and C/C genotype followed.Significant differences were found among these 3 groups.Conclusion FK506's blood concentration after orally application is associated with the mdr1 gene C3435T polymorphism.Detecting the polymorphism of mdr1 gene C3435T is helpful when FK506 is given orally. |
