新型基因工程人肿瘤坏死因子联合足叶乙甙治疗小鼠肺癌的实验研究

摘    要:目的 探讨新型基因工程人肿瘤坏死因子(nrh T N F) 和足叶乙甙( V P- 16) 的协同抗肿瘤作用及nrh T N F 的毒副作用。方法 以复制成功的 Lewis 肺癌(3 L L) 小鼠为模型局部用药。结果 nrh T N F 或 V P- 16 瘤体内注射均能使肿瘤生长及肺转移受到一定的抑制,抑瘤率分别为3371 % 、3046 % ,肺转移瘤数与对照组比较 P < 005 ,肿瘤出现一定程度坏死。而联合用药则能显著抑制肿瘤生长及肺转移,抑瘤率为6577 % ,大于理论抑瘤率5391 % ,肺转移瘤数与对照组比较 P < 001 ,肿瘤出现广泛的出血坏死。同时观察到nrh T N F 对实验小鼠无明显毒副作用。结论 nrh T N F 和 V P- 16联合应用,具有协同抗肿瘤作用,对于肺癌治疗具有进一步临床应用探讨价值。目的 探讨新型基因工程人肿瘤坏死因子(nrh T N F) 和足叶乙甙( V P- 16) 的协同抗肿瘤作用及nrh T N F 的毒副作用。方法 以复制成功的 Lewis 肺癌(3 L L) 小鼠为模型局部用药。结果 nrh T N F 或 V P- 16 瘤体内注射均能使肿瘤生长及肺转移受到一定的抑制,抑瘤率分别为3371 % 、30

Therapeutic effect of new recombinant human tumor necrosis factor combined with etoposide on m urine lung carcincoma
Abstract:Objective To investigate the effect of new recombinant human tumor necrosisfactor(nrh T N F) combined with etoposide( V P- 16) on inhibition of murine lung cancer and the side effect of T N Fin experimental mice . Methods Tumor- bearing mice wererandomly divided into four groups ,normalsaline ,nrh T N F, V P- 16 and nrh T N Fplus V P- 16 were injected into the neoplasm ofthe fourgroups rospectively . Four mice of each group were killed 14 days afterinoculation after,neoplasms were detected underlight microscope ;others were killed after 21 days inoculation ,neoplasms were weighted ,and lung metastatic tumors were counted . Results Both nrh T N Fand V P- 16 inhibited the growth and lung metastasis of murine 3 L Ltumor,inhibition ratesin nrh T N Fgroup and V P- 16 group were 33 .71 % and 30 .46 % ,respectively ,and tumor necrosis resulted to a certain degree . The lung metastatic tumors in the groups mentionedabove were fewer than in control group ( P < 0 05) . Afavourable out- come was observed in the animals treated with nrh T N F/ V P-16 ,i.e .lower tumor growth rate (inhibition rate 65 .77 % ) and more extensivetumor hemonagic necrosis oppeareathan in control group( P < 0 01) .nrh T N Fprodtced no marked side ef
