p21Ras对哮喘大鼠气道平滑肌细胞增殖的影响

摘    要:目的观察p21Ras对哮喘大鼠气道平滑肌细胞增殖的影响。方法 14只SD大鼠,随机分为哮喘组和对照组,建立大鼠哮喘模型,采用酶消化法培养气道平滑肌细胞,免疫组织化学鉴定为平滑肌细胞,透射电镜观察细胞形态,分别取第3代对照组及哮喘组细胞,各自分为以下4个处理组,即对照组分为:a.对照空白组:不加任何干预剂;b.对照干预组;哮喘组分为:c.哮喘空白组;d.哮喘干预组。加入10mMFPTIII干预,流式细胞仪分析细胞周期分布,经Lev-ene’s方差齐性检验,按α=0.05水准,4组总体方差齐,组间比较采用Bonferron法检验,结果通过p21Ras干预后,哮喘空白组G0/G1期气道平滑肌细胞细胞(AMSC)比例显著低于对照空白组及哮喘干预组(P0.05);S期和S+G2/M期ASMC比例显著高于上述两组(P0.05);对照空白组与对照干预组差别不显著(P﹤0.05)。结论 p21Ras影响哮喘大鼠气道平滑肌细胞周期时相中G0/G1的比例,使其增殖受到抑制,提示p21Ras与哮喘气道平滑肌的增殖密切相关。

Abstract:Objective To investigate the effect of p21Ras on the proliferation of airway smooth muscle cell of rats. Methods The 14 SD rats were randomly assigned into two groups: asthmatic group and control group. Rat model of asthma was established, using enzymatic digestion and cultured airway smooth muscle cells, ASMCs were identified by immunohistoehemistry, and ultrastructure of cells were compared by electron microscopy; third generation ASMC of both groups were taken, then divided into four treatment groups: a. blank control group without any intervention agents; b. control intervention group; c. asthma blank, group; d.asthma intervention group. 10mMFPTIII was added and cell cycle distribution was analyzed by flow cytometric. Homogeneity of variance among four groups were compared using Bonferron test method in accordance with Levene's homogeneity of variance test and et = 0.05 level.Results After p21Ras intervention, airway smooth muscle ceils (AMSCs) in rats of asthma blank group at G0/G1 phase was significantly lower than that of the control blank group and asthma intervention group I (P 〈0.05); ASMCs at S phase and S + G2 / M phase was significantly higher than in the two above-mentioned groups (P 〈0.05); no significant difference was found between blank control group and intervention group I (P〉 0.05). Conclusion p21Ras affect cell cycle at G0/G1 phase proportion of airway smooth muscle from asthmatic rat, and proliferation was inhibited, suggesting p21Ras was closely related to airway smooth muscle proliferation in rats.
