灯盏花素对糖尿病肾病大鼠肾组织MMP-9表达的影响

目的观察灯盏花素对链脲佐菌素(STZ)诱导糖尿病肾病(DN)大鼠肾组织基质金属蛋白酶(MMP)-9表达的影响。方法 STZ诱导的糖尿病SD大鼠模型,雌性SD大鼠随机分为正常对照组(N)、DN模型组(DN)、DN模型+灯盏花素治疗组(DS)三组。模型建立后,在第12周,测量血糖、24 h尿蛋白定量;12周末处死大鼠,检测尿素氮(BUN)、血肌酐(Scr);取大鼠肾组织行HE染色,进行病理组织学观察;用免疫组化法检测肾组织MMP-9。结果实验12周末,与DN模型组比较,DS组大鼠24 h尿蛋白定量、BUN、Scr显著降低(P0.05);肾组织病理改变减轻,免疫组化结果显示,肾组织MMP-9明显增加(P0.05)。结论灯盏花素可增加MMP-9表达,降低蛋白尿,保护肾脏功能。提示灯盏花素可能通过调节MMP-9的表达,从而减缓糖尿病肾病的发生和发展。

Effect of breviscapine on expression of renal tissue MMP-9 in rats with diabetic nephropathy

Objective To observe the effect of breviscapine on the expression of MMP-9 in renal tissue of streptozotocininduced diabetic nephropathy rats.Methods The diabetic SD rats were induced by STZ, female SD rats were divided intonormal control group（ N group）, Diabetic Nephropathy group（ DN group）, diabetic breviscapine treatment group（ DS group）.After construction of the model he levels of blood glucose（ Glu） and the 24 h urinary protein were determined.After 12 weeksthe rats were killed and the levels of serum creatinine（ Scr） and urea nitrogen（ BUN） in each groups were determined. Kidneytissues stained with HE was histopathologically observed and the expression of MMP- 9 in renal tissue was also detected.Results After 12w,the 24 h urinary protein,BUN and Scr in the DS group,compared with DN group,were significantlyreduced（ P 0.05）,the expression levels of MMP-9 were significantly increased（P〈0.05）.Conclusion Breviscapine canenhance the expression of MMP-9, reduce proteinuria and protect renal function,suggesting that breviscapine could regulatethe expression of MMP-9 and delay the occurrence and development of diabetic nephropathy.
