用McAb检测患者血清循环抗原诊断黑热病的研究

作者首次报道应用单克隆抗体(McAb)检测黑热病患者血清内循环抗原获得成功。发现硝化纤维滤膜可以较好地吸附该循环抗原,再与高滴度McAb C-2结合及 ELISA显色(McAb-AST)可取得很好的效果。对20例确诊黑热病患者血清的试验结果,19例呈阳性反应,阳性率95％。正常人对照、间日疟及吸虫患者均显阴性反应。检测结果说明本试验的敏感性、特异性及重复性都很好,方法简易可行,对我国黑热病的诊断具有较好的前景。

DETECTING L. DONOVANI CIRCULATING ANTIGEN BY McAb IN THE DIAGNOSIS OF KALA-AZAR

Immune diagnosis of kala-azar has been based on detecting specific antibodies in the patients. Detecting L. donovani antigen of the infected subjects, of which there has been no report yet, would serve as an alter-native. Using anti-L. donovani McAb C-2produced in this laboratory, we have con-ducted McAb-AST for detecting the circu-lating antigen in kala-azar cases. Of the 20 sera tested, 19 showed positive findings.while sera of 51 normal controls all gave negative results. When the color-reaction of the McAb-AST was subjected to OD-scan-ning, it was found that the difference between the kala-azar patients group and the normal controls was highly significant (t test, P<0. 001). No crossreactions were observed when sera of 22 malaria, 11 par- agonimiasis and schistosomiasis cases were tested in the same way. Our studies showed that the McAb-AST is a sensitive and specific assay relatively simple to conduct,and it is apparently a useful tool in diagno-sing visceral leishmaniasis.
