调肠消炎片调控溃疡性结肠炎大鼠模式识别受体TLR-4信号通路的研究

摘    要:目的:观察调肠消炎片对溃疡性结肠炎(UC)大鼠结肠的炎症反应及血清toll样受体4(TLR-4)浓度的影响。方法:用复合法(2,4,6-三硝基苯磺酸加乙醇)造模制备UC大鼠模型,将60只SD大鼠随机分为正常组、模型组、调肠消炎片组及柳氮磺胺吡啶(SASP)组,除正常组外,其余各组从造模后第3天开始分别每天灌胃给药1次,灌胃14天后取材观察大鼠结肠炎症反应情况及血清TLR-4浓度。结果:模型组CMDI及血清TLR-4浓度均高于正常组(P<0.01)。调肠消炎片组的CMDI及血清TLR-4浓度均低于模型组(P<0.05,P<0.01)。结论:调肠消炎片明显降低UC大鼠血清TLR-4浓度,提示其可能通过调控TLR-4信号通路,有效治疗溃疡性结肠炎模型大鼠。

Effect of Tiaochang Xiaoyan Tablets on Pattern Recognition Receptor TLR-4 Signal Pathway in Rats with Ulcerative Colitis

Abstract:Objective:To investigate the effects of Tiaochang Xiaoyan Tablets(TXT) on the inflammatory reaction and serum concentration of Toll-like receptor(TLR-4) in the rats with ulcerative colitis(UC).Methods:Ulcerative colitis was induced in rats by giving the enema of trinitrobenzene sulphonic acid(TNBS) and ethanol.Sixty rats were randomized into 4 groups: normal group,model group,sulfasalazine(SASP) group and TXT group.Except the normal group,the other groups were given intragastrically normal saline,SASP and TXT,respectively.The medication was from the 3rd day to the 17th day after the model was established.At the end of the experiment,the degree of the colonic inflammation and the serum concentration of TLR-4 were examined.Results:Compared with the normal group,the colon mucosal inflammation index(CMDI) and the concentration of TLR-4 in the model group were significantly up-regulated(P < 0.01).Compared with the model group,the CMDI and the concentration of TLR-4 in the TXT group was significantly down-regulated(P < 0.05 or P < 0.01).Conclusion:TXT exert effects on down-regulating TLR-4 expression in ulcerative colitis rats,and its mechanism is probably related with the regulation of TLR-4 signal pathway.
