重组补体抑制剂TP10对大鼠脊髓损伤组织中性粒细胞浸润的影响

摘    要:目的探讨重组强效补体抑制剂TP10对大鼠脊髓损伤组织中性粒细胞浸润程度及脊髓损伤后神经功能的影响。方法制备大鼠急性脊髓损伤模型,观察伤后6,12,24h、3,7d时间点TP10组与对照组大鼠脊髓损伤组织中性粒细胞浸润情况、补体C9阳性表达的部位及时程;测定髓过氧化物酶(MPO)活性;采用BBB评分法评定大鼠后肢运动功能。结果TP10组在伤后各时间点中性粒细胞浸润程度均明显轻于对照组;TP10组在伤后各个时间点C9阳性表达均明显轻于对照组,差异有极显著性(P〈0.01);TP10组在伤后各时间点损伤组织MPO活性弱于对照组,差异有极显著性(P〈0.01);TP10组在伤后3、7d时间点大鼠后肢BBB评分明显优于对照组(P〈0.05)。结论重组强效补体抑制剂TP10可通过抑制补体系统激活机制显著减轻急性脊髓损伤组织的中性粒细胞浸润,对脊髓损伤有保护作用。

Abstract:Objective To determine the effects of strengthened complement inhibitor TP10 on the degree of neutrophil infiltration in spinal cord injury in rats and on the function of nerve after spinal cord injury. Methods Neutrophil infiltration and the expression sites and time course of C9 were observed,Myeloperoxidase activity were assessed in the spinal cord followling acute injury,motor function of the rat hind limbs were evaluated by the the BBB scores at 6 h,12 h,24 h,3 d and 7 d after injury in TP10 group and control group. Results Neutrophil infiltration in TP10 group at each time point after injury was obviously less than that in control group. The positive expression of C9 in TP10 group at each time point after injury was obviously less than that in control group with highly significant difference (P < 0.01). Myeloperoxidase activity in TP10 group at each time point after injury was obviously less than that in control group with highly significant difference (P <0.01). BBB scores in TP10 group at 3,7d was better than that in control group(P < 0.05). Conclusion Strengthened complement inhibitor TP10 can lessen the neutrophil infiltration in acute spinal cord injury by inhibiting the activation of the complement system and have protective effect on spinal cord injury.
