基于网络药理学的人参白虎汤治疗糖尿病药效机制研究

摘    要:目的利用网络药理学的方法对人参白虎汤在治疗糖尿病的过程中药效机制进行研究。方法在中药系统药理学分析平台(TCMSP)中得到人参白虎汤中知母、甘草和人参的化学成分和药物靶点,在BATMAN-TCM中得到粳米的化学成分和药物靶点,在HTDocking中得到石膏的靶点;利用在线人类孟德尔遗传数据库(OMIM),人类基因数据库(GeneCards)筛选糖尿病的相关靶点;运用Cytoscape 3.7.1构建“化合物—靶点—基因”网络;运用蛋白相互作用(PPI)网络筛选出核心靶点;运用蛋白相互作用(PPI)网络筛选出核心靶点;通过生物学信息注释数据库(DAVID)进行基因本体(GO)功能富集分析和基于京都基因与基因组百科全书(KEGG)通路富集分析。结果构建出化合物靶点网络,蛋白质相互作用网络,对核心蛋白进行基因本体功能分析和通路分析。结论本实验利用网络药理学对人参白虎汤和其治疗的糖尿病进行了分析,探究其共性作用机制及高频共性靶点,高频共性基因,高频共性GO功能和高频共性KEGG通路等,为人参白虎汤用于糖尿病的治疗提供理论参考。

Abstract:Objective To explore the pharmacodynamic mechanism of Renshen Baihu Decoction on the treatment of diabetes mellitus based on network pharmacology techniques.Methods The chemical components and drug targets of Zhi Mu,Gan Cao and Ren Shen in Renshen Baihu Decoction were obtained in Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP).The chemical components and drug targets of japonica rice(Jing Mi)were obtained in Bioinformatics Analysis Tool for Molecular mechANism of Traditional Chinese Medicine(BATMAN-TCM),and the target of gypsum(Shi Gao)was obtained in HTDocking.The Online Mendelian Inheritance in Man(OMIM)database and the GeneCards database(GeneCards)were used to screen diabetes related targets.Cytoscape3.7.1 software was used to construct“chemical-target-gene”network.Protein-protein interaction(PPI)network was used to screen out the core targets.Gene ontology(GO)functional enrichment analysis and Kyoto encyclopedia of genes and genomes(KEGG)pathway enrichment analysis were performed by Database for Annotation,Visualization and Integration Discovery(DAVID).Results Compound target network and PPI network were constructed,and GO function analysis and pathway analysis of core proteins were performed.Conclusion Based on network pharmacology,the mechanism of Renshen Baihu Decoction on treating diabetes is explored by performing common mechanism of action,high-frequency common targets,high-frequency common GO function and high-frequency common KEGG pathways,which may provide theoretical basis for future research.
