弓形虫致密颗粒蛋白的免疫反应性评价

摘    要:目的对弓形虫的重组蛋白谷胱甘肽巯基转移酶致密颗粒蛋白(GSTGRA7)的免疫反应性进行评价。方法异丙基βD硫代半乳糖苷(IPTG)诱导大肠埃希菌BL21(pGEX4T1/GRA7)表达,表达产物进行十二烷基硫酸钠(SDS)变性蛋白质电泳,分别以人抗弓形虫阳性血清和兔抗弓形虫阳性血清为一抗进行WesternBlotting分析。用GST亲和层析柱纯化重组蛋白,以此蛋白作为包被抗原,酶联免疫吸附试验(ELISA)检测抗弓形虫阴性、阳性血清。结果重组蛋白GSTGRA7能被兔抗弓形虫阳性血清、人抗弓形虫阳性血清所识别。该蛋白能与人抗弓形虫阳性血清、兔抗弓形虫阳性血清特异结合,而与抗弓形虫阴性血清无反应。结论弓形虫重组蛋白GSTGRA7具有良好的免疫反应性。

Abstract:Objective To evaluate on the immunoreactivity of the recombinant protein GST-GRA7 of toxoplasma gondii.Methods The Escherichia coli.BL21,which had recombinant plasmid pGEX-4T-1/GRA7, was induced by IPTG. The protein was analyzed by SDS-PAGE and Western blotting with positive sera. .Then the protein was purified and used as a diagnostic antigen for ELISA to test positive sera and negative sera. Results The protein could react with human/rabbit positive sera analyzed by Western blotting. Conclusion The recombinant protein GST-GRA7 has very good immunoreactivity.
