谷氨酰胺对饥饿大鼠肠黏膜免疫系统中CD4~+、CD8~+及SIgA表达的影响

摘    要:目的:观察谷氨酰胺对饥饿大鼠肠黏膜免疫学指标的影响。方法:以饥饿SD大鼠为模型,将90只雄性SD大鼠随机分成三组(即正常组、饥饿组、谷氨酰胺组)。采用饥饿大鼠模型,运用免疫组织化学染色法标记小肠黏膜中CD4+、CD8+T淋巴细胞及SIgA数量,用图像分析仪统计其含量。结果:与正常组比较,全饥饿组大鼠肠黏膜免疫系统中SIgA及CD4+、CD8+T淋巴细胞数量明显减少,差异有统计学意义(P<0.05)。补充谷氨酰胺(GLN)后,SIgA、CD4+、CD8+T淋巴细胞数量明显升高,补充谷氨酰胺组与全饥饿组比较差异有统计学意义(P<0.01)。补充谷氨酰胺组与正常组比较,SIgA数量在饥饿后3 d、7 d无差异,而在饥饿后5 d、9 d差异有统计学意义(P<0.01);CD8+T淋巴细胞数量在饥饿后各时间点明显减少,差异有统计学意义(P<0.01);CD4+T淋巴细胞数量在饥饿后3 d、5 d未见有明显差异,在饥饿后7 d、9 d则明显减少,差异有统计学意义(P<0.01)。结论:谷氨酰胺可增强饥饿大鼠肠黏膜的免疫功能,发挥肠黏膜的保护作用。

Abstract:Objective To observe the effect of glutamine on immunological indexes in intestinal mucosal immune system of complete starved rats.Methods 90 male SD rats were randomly divided into 3 groups:normal control,starvation and glutamine.CD4+,CD8+T-LYM and SIgA in intestine of rat were labeled by immunohistochemical staining and were measured using imaging analyzer.Results Compared with the normal group,the numbers of CD4+,CD8+T-LYM and SIgA in starved rat reduced significantly(P0.05).After treatment with GLN,the numbers of CD4+,CD8+T-LYM and SIgA increased significantly(P0.01).Compared with the normal group,in the GLN group the numbers of SIgA had significant difference at 5,9 days after starvation(P0.01)and the numbers of CD8+T-LYM were much lower at all phases after starvation(P0.01)and the numbers of CD4+T-LYM were much lower at 7,9 days after starvation(P0.01).Conclusion Glutamine is helpful to increase immune function of intestinal mucosa and can alleviate the damage of intestinal mucosa after starvation.
