淋球菌分离株耐药质粒谱型研究

摘    要:目的了解淋球菌临床分离株的质粒分布情况,初步探讨其与淋球菌耐药性的关系。方法应用碱裂解法对淋球菌的质粒进行抽提,获得淋球菌临床分离株的质粒谱。结果共检测出有42.5kb,39.5kb,7.4kb和4.2kb的四种不同分子量的质粒,7.4kb和4.2kb质粒检出率分别为75.00%和96.25%。质粒谱型有10种,其中以42.5kb+39.5kb+7.4kb+4.2kb,39.5kb+7.4kb+4.2kb,7.4kb+4.2kb及4.2kb四种类型居多,占86.25%。所有检测出的PPNG菌株(包括PPNG/TRNG)均含有7.4kb质粒,所有TRNG菌株(包括PPNG/TRNG)均含有42.5kb质粒和39.5kb质粒;而非PPNG或TRNG中所携带的质粒谱型则相对复杂。结论7.4kb的质粒可能会介导淋球菌高水平的青霉素耐药性,42.5kb及39.5kb质粒可能与介导四环素高水平耐药有关。

Abstract:Objective To study the distribution of plasmids in the clinical Neisseria gonorrhoeae isolates,and explore the relation between plasmids and drug resistance.Methods The plasmid profiles were obtained by using alkaline-lysis plasmid extraction method.Results A total of four types of plasmid were detected (42.5 kb,39.5kb,7.4 kb and 4.2 kb),among them,the percentages of presence of 7.4 kb and 4.2 kb plasmids were 75.00% and 96.25%,respectively.A total of ten types of plasmid profiles were observed.Four main types were 42.5 kb 39.5 kb 7.4 kb 4.2 kb type,39.5 kb 7.4 kb 4.2 kb type,7.4 kb 4.2 kb type and 4.2 kb type,accounting for 86.25% of total isolates.All the PPNG strains carried a 7.4 kb plasmid,all the TRNG strains carried both 42.5 kb and 39.5 kb plasmids,while the plasmid profile of non PPNG or TRNG varied greatly.Conclusion The results indicated that the 7.4 kb plasmid might mediate high level penicillin resistance,while the 42.5 kb and 39.5 kb plasmids might have relationship with tetracycline resistance.
