摘    要:摘要目的研究两面针中木脂素化合物结晶8(简称结晶8)对甲醛所致伤害性疼痛大鼠的即早基因(c fos)表达的影响。方法SD雌性大鼠34只,随机分为5组:0.9%氯化钠溶液组、模型组、阳性对照组(罗通定注射液2 mg·kg-1)、结晶 8高剂量(2 mg·kg-1)组各7只,结晶 8低剂量(1 mg·kg-1)组6只。采用免疫组化染色观察疼痛大鼠c fos基因的表达情况。结果模型组大鼠脑干、下丘脑及脊髓视野中Fos阳性颗粒分别为(20.33±2.63),(19.28±2.42),(18.36±5.11)颗;结晶 8高剂量组分别为(10.76±3.66),(12.49±2.80),(6.87±2.10)颗。结论结晶 8的镇痛机制可能与抑制中枢FOS蛋白的表达有关。

Abstract:Objective To study the effect of the crystal-8 from zanthoxylum nitidum(Roxb.) DC.(crystal-8) on the expression of c-fos gene in rats with nociceptive pain caused by formalin.Methods A total of 34 female SD rats were randomly divided into 5 groups:the blank control,model control,positive control(rotundine 2 mg·kg-1),high dose of crystal-8 groups(2 mg·kg-1),7 rats in each,and 6 rats in the low dose of crystal-8 group(1 mg·kg-1).The expression of c-fos gene in painful rats was detected by using immunohistochemistry.Results The positive particles of FOS protein in the brainstem,hypothalamic and spinal cord of the model control group were(20.33±2.63),(19.28±2.42),(18.36±5.11),respectively,those in the high dose of crystal-8 group was(10.76±3.66),(12.49±2.80),(6.87±2.10),respectively.Conclusion The mechanism of analgesia by crystal-8 may be related to restraining the FOS protein expression in central nervous tissues.
