摘    要:目的为青牛胆的物种鉴定和遗传图谱的构建寻找一种新的途径。方法采用序列相关扩增多态性(Sequence related amplified polymorphism,SRAP)技术对青牛胆DNA进行PCR扩增,逐级优化反应参数。结果优化得到稳定重复性好的青牛胆SRAP反应体系。结论SRAP技术在分子水平上对青牛胆进行鉴定是一种行之有效的手段,为今后进一步的青牛胆物种鉴定、遗传图谱的构建等研究奠定基础。

Abstract:Objective To develop a new method for Tinospora sagittata species identification and genetic map construction. Methods Sequence-related amplified polymorphism (SRAP) was applied for T. sagittata to carry on PCR amplification of its DNA and optimize the reaction parameter grade by grade. Results The stable and reproducible SRAP reaction system of T. sagittata has been developed. Conclusion SRAP is an effective method for T. sagittata identification in molecular degree and it has set up a foundation for the further species identification and genetic map construction of T. sagittata.
