非小细胞肺癌与淋巴结转移癌组织中FHIT基因异常表达的相关性

摘    要:目的 :检测脆性组氨酸三联体 (FHIT)基因在正常肺组织、非小细胞肺癌 (non smallcelllungcancer)与淋巴结转移癌组织中表达 ,探讨FHIT基因与肺癌发生及转移的关系。方法 :用免疫组化SP(streptavidinperoxidaseconju gatedmethod ,SP)法检测 2 0例无淋巴结转移的肺癌组织 ,2 0例肺癌及其相应的淋巴结转移癌组织和 15例正常肺组织中FHIT基因的表达水平 ,用图像分析系统测量FHIT蛋白表达的阳性率和平均光密度。结果 :FHIT蛋白在正常肺组织、无淋巴结转移的肺癌、肺癌及其相应的淋巴结转移癌组织中的阳性率分别为 89.1%、6 0 .5 %、5 4 .1%、30 .4 % ,平均光密度分别为 (0 .2 74± 0 .0 2 3)、(0 .16 5± 0 .0 16 )、(0 .0 87± 0 .0 0 4 )、(0 .0 4 9± 0 .0 0 7)。肺癌组织FHIT蛋白表达阳性率和强度明显低于正常肺组织 (P <0 .0 1) ,淋巴结转移癌组织FHIT蛋白表达明显低于其它 3组 (P <0 .0 1)。结论 :在肺癌发生、浸润、转移病变阶段中 ,FHIT基因表达下调 ,这揭示肺癌的演进过程与FHIT有密切关系。

Abstract:Objective: To investigate abnormal expression of the fragile histidine triad gene in non small cell lung cancer and the role of FHIT gene in metastasis. Methods: Immunostainings were utilized to measure levels of protein expression of FHIT in following four study groups: 15 normal lung samples,20 lung cancer samples without metastasis,20 lung cancer samples with metastasis and 20 matched metastatichilar lymphnodes of lung cancer patients. The positive rate and average light density value of FHIT protein expression were measured by guantitatove image analysis. Results: FHIT expression of protein in patients with lung cancers was lower significantly than that with normal lung. The positive rate of FHIT protein was 89.05% vs 60.45%,54.10%,30.36%,average light density value was (0.274±0.023)vs(0.165±0.016),(0.087±0.004),(0.049±0.007). Successively the expression of FHIT in metastatichilar lymphnodes was lower significantly than that of normal lung samples, lung cancer samples without metastasis. The expression in lung cancer with metastasis was lower significantly than that in lung cancer without metastasis (P<0.01). Conclusion: These data indicate that reduction of FHIT gene expression is frequent in lung cancer. FHIT may play an important role in the progression and metastasis of lung carcinoma.
