菱角纯化物三羟基苯甲酸二聚体对肝癌SMMC-7721细胞凋亡及细胞周期进程的影响

摘    要:目的:探讨菱角纯化物3,4,5-三羟基苯甲酸二聚体对肿瘤生长抑制的可能机制。方法:应用流式细胞术检测体外培养人肝癌SMMC-7721细胞凋亡及细胞周期进程的影响。结果:不同剂量(3.125、6.250、12.500和25.000 mg?L-1)菱角纯化物分别处理细胞后,其凋亡百分数均明显高于对照组(P<0.001)。G2+M期细胞百分数增高,其中3.125、6.250和12.500 mg?L-1菱角纯化物组明显高于对照组(P<0.05或P<0.01);S期细胞百分数低于对照组,尤其是6.250、12.500和25.000 mg?L-1纯化物组降低明显(P<0.05)。结论:菱角纯化物3,4,5-三羟基苯甲酸二聚体对肿瘤生长的抑制作用可能通过G2+M期阻滞引起细胞凋亡发挥作用。

Abstract:Objective To observe the effects of purified product trihydroxybenzoic acid dimmer in different doses from Trapa manshurica Fler on apoptosis and cell cycle progression of liver cancer SMMC-7721 cells in vitro and explore its possible mechanism of inhibiting tumor growth. Methods The changes of apoptosis and cell cycle progression were measured with flow cytometry. Results The apoptotic percentages of the cells treated with the purified products with doses of 3.125, 6.250, 12.500 and 25.000 mg·L~ -1 increased significantly as compared with that in the control (P<0.001). And the percentages of G_2+M phase cells increased, those in 3.125, 6.250 and 12.500 mg·L~ -1 groups were significantly higher than that in the control (P<0.01 or P<0.05); while the percentages of S phase cells decreased,especially decreased in 6.250, 12.500 and 25.000 mg·L~ -1 group(P<0.05). Conclusion The inhibitory effect of purified product trihydroxybenzoic acid dimmer from Trapa manshurica Fler on tumor growth could be due to cell apoptosis caused by G_2+M phase arrest.
