Abstract:Objective: To investigate the effect of interleukin-6 (IL-6) antisense oligonucleotide (ASODN) on proliferation and apoptosis of human bileductcarcinoma cell line HCCC-9810. Methods: IL-6 ASODN was transfected into HCCC-9810 cells by LipofectamineTM 2000. Cell colony efficiency experiment and MTT were adopted to determine HCCC-9810 cells’ proliferation. The apoptosis was analyzed by flow cytometry. The expression of IL-6 mRNA was detected by RT-PCR. Results: After transfection, the cell clone forming rate in the ASODN transfected cells was (4.58±0.35)%, which was significantly lower than that in the control cells (P<0.05). The cell proliferation inhibition rate in the ASODN transfected cells was 67.2%, which was significantly higher than that in the control cells (P<0.05). The apoptosis rate in the ASODN transfected cells was (10.25±0.5) %, which was significantly higher in the ASODN transfected cells than that in the control cells (P<0.05). The IL-6 mRNA was decreased by 60.52% in the ASODN transfected cells as compared with that in the blank group, and was significantly lower than that in the control cells (P<0.05). Conclusion: ASODN targeting at IL-6 can inhibit HCCC-9810 cells’ proliferation and induce apoptosis.
