摘    要:目的 探讨芍药甙(PAE)对PC12细胞缺血样损伤模型的保护作用及其机制。方法 采用组织培养法,以肾上腺嗜铬瘤克隆化细胞株PC12细胞为材料,制备缺糖损伤、缺氧损伤、自由基损伤Caf损伤、NO损伤及GIu毒性损伤模型。结果 形态学检查发现PAE对6种脑缺血样损伤模型中PC12细胞具有明显保护作用,MTT染色提示PAE可显著提高损伤模型中PC12细胞的存活数,降低胞浆释放的LDH水平。结论 除缺氧损

Abstract:To investigate protective effect and mechanism of paeoniflorin(PAE)on ischemia- like injury models. Methods:Six ischemic injury models including hypoxia,hypoglucose,oxidant injury,calcium overload,nitric oxide neurotoxicity and glutamic acid injury were used to assay the anti- ischemic effect of PAE in cultured PC12 cells. Results:The morphological examination founded that PAE had obvious protective effect on PC12 cells in above six models.MTT staining and LDH measurement also indicated that PAE significantly increased number of live cells and lessened the extent of cell injury. Conclusion:PAE protected PC12 cells from ischemic injury effectively in vitro.The mechanism of its action may focus on blocking calcium channel of cell membrane.
