高良姜素对PC12细胞氧化损伤的保护作用及其机制研究

摘    要:目的:探讨高良姜素对H_2O_2诱导的PC12细胞氧化损伤的保护作用及其作用机制。方法:用H_2O_2损伤PC12细胞建立氧化应激损伤模型,细胞分为正常对照组、模型对照组、高良姜素(低、中、高浓度)组,并给予相应干预措施。采用MTT法检测细胞生长抑制率,采用Annexin-V-FITC凋亡检测试剂盒检测细胞凋亡情况,免疫印迹法检测蛋白表达情况。结果:高良姜素预处理可以减少H_2O_2诱导的PC12细胞氧化应激损伤;高良姜素能明显抑制细胞凋亡,且呈剂量依赖性;高良姜素可以增加PI3K/Akt和Bcl-2家族蛋白比例。结论:高良姜素是通过激活PI3K/Akt信号通路发挥对H_2O_2诱导的PC12细胞氧化损伤的保护作用。

Abstract:  Objective:To study the protective effect and its possible mechanisms of galangin on H2O2-induced cytotoxicity in PC 12 cell.   Methods: A model of oxidative stress in PC12 cells injury was established by H2O2.Cells were divided into control group, model group, galangin(low-, medium-, high-dosage)groups. The viability of PC12 cells was assessed by MTT assay, cell apoptosis was detected by Annexin-V-FITC Apoptosis Detection Kit, and Western blotting method was carried out to detect the protein expressions.   Results: Pretreatment with galangin could reduce H2O2-induced oxidative stress; galangin significantly inhibited cell apoptosis with a dose-dependent manner; and galangin could increase the PI3K/Akt and Bcl-2 family prote in ratios in cells.   Conclusion: Galangin can protect H2O2-induced PC 12 cell injury by activating PI3K/Akt pathway.
