加味四君子汤对脾气虚肺癌转移小鼠Fas和Bcl-2 mRNA表达的影响

摘    要:目的:探讨脾气虚对肺癌转移的影响机制以及加味四君子汤的作用机制。方法:采用苦寒泻下、劳倦、饮食节制复合因素法建立脾气虚小鼠模型,连续16 d。造模第17天眼球取血测血清胃泌素浓度,综合验证脾气虚模型。脾气虚模型成功后C57小鼠右腋下接种Lewis肺癌细胞,制备肺癌小鼠模型。小鼠随机分为正常组、脾气虚模型组、肺癌组、脾气虚肺癌组、脾气虚肺癌健脾组,肺癌组及脾气虚肺癌组生理盐水灌胃,脾气虚肺癌健脾组加味四君子汤灌胃。14 d后观测肺转移结节数,采用Real-time PCR方法检测Fas、Bcl-2基因的mRNA表达。结果:与脾气虚肺癌组比较,肺癌组、脾气虚肺癌健脾组肺转移结节均较少;与肺癌组比较,脾气虚肺癌组Fas mRNA表达下调,Bcl-2 mRNA表达上调;与脾气虚肺癌组比较,脾气虚肺癌健脾组Fas mRNA表达上调,Bcl-2 mRNA表达下调。结论:脾气虚可通过抑制Fas mRNA表达及促进Bcl-2 mRNA表达,促进肺癌转移;加味四君子汤可通过促进Fas mRNA表达及抑制Bcl-2 mRNA表达,抑制肺癌转移。

Abstract:  Objective:To investigate the influence of spleen and qi deficiency on the metastasis of lung cancer, and the mechanism of modified Sijunzi decoction on lung cancer metastasis with spleen and qi deficiency.   Methods: Model of spleen and qi deficiency was established in mice by multifactor methods of cold and bitter,purgation, fatigue and food intake restriction, for continuous 16 days. On the 17th day, the blood were collected and serum concentration of gastrin was detected,which verified the model of spleen and qi deficiency. Lewis lung cancer cells were subcutaneously injected into the right armpit of successful modeling mice to establish the lung cancer model. Mice were divided into the normal group,spleen and qi deficiency model group,lung cancer group,lung cancer with spleen and qi deficiency group and modified Sijunzi group. The lung cancer group and lung cancer with spleen and qi deficiency group were administrated orally with normal saline, the modified Sijunzi group was administrated orally with modified Sijunzi decoction. After treatment for 14 days,the numbers of the lung metastatic nodules were counted,the gene expression of Fas and Bcl-2 were detected by Real-time PCR.   Results: The metastatic nodules of the lung cancer group and modified Sijunzi group were less than those of the lung cancer with spleen and qi deficiency group. Compared with the lung cancer group,the Fas mRNA expression was down-regulated and the Bcl-2 mRNA expression was up-regulated in the lung cancer with spleen and qi deficiency group. Compared with the lung cancer with spleen and qi deficiency group,the Fas mRNA expression was up-regulated and the Bcl-2 mRNA expression was down-regulated in the modified Sijunzi group.   Conclusion: Spleen and qi deficiency could down-regulate the expression of Fas mRNA and up-regulate the expression of Bcl-2 mRNA,which promoted the metastasis of lung cancer. Modified Sijunzi decoction could up-regulate the expression of Fas mRNA and down-regulate the expression of Bcl-2 mRNA, which inhibited the metastasis of lung cancer.
