人肺癌组织一氧化氮合酶及总RNA表达水平的梯度改变

摘    要:目的:探讨一氧化氮合酶(NOS)在肺癌组织中的表达与作用机制。方法:按自身配对原则收集人肺癌癌灶组织、癌灶周围组织及远离癌灶的肺组织各26份,作病理组织学检查并制备肺组织匀浆,分析不同肺组织中总癌周围组织>远离癌灶肺组织的梯度分布;癌组均显著高于癌周及远癌组织(P<0.001),癌组织中总RNA水平与NOS浓度呈显著正相关(P<0.01);鳞癌组与腺癌组间总RNA和NOS的表达水平未见明显差异(P>0.05);分化程度高的肺癌组NOS表达水平较中分化和低分化肺癌组低;癌灶组织中NO浓度明显高于远癌组织(P<0.05)。结论:肺癌组织中RNA的高度表达说明肿瘤组织中蛋白质合成旺盛,而NOS的过度表达则可能与加速生成NO以杀伤肿瘤细胞有关。

Abstract:Objective:To explore the expression and alteration mechanism of nitric oxide synthetase (NOS) in tissues of lung cancer.Methods: Human lung cancers, paracancerous and distant cancerous tissues with self-pair control were collected, homogenates were prepared, the concentrations of total RNA, NOS and nitric oxide (NO) in 26 patients with lung cancer were investigated in the present study. Results: The expression intensity of lung tissue total RNA and NOS showed grade distribution from lung cancerous tissues to distal cancerous tissues, they are significantly higher in lung cancer than those of its paracancerous tissues or its distant cancerous ones. The positive relationship (P<0.01) was found between total RNA and NOS in lung cancer tissues. No significantly difference (P>0.05)of total RNA or NOS was found between adenocancinoma group and squamous cell carcinoma group.The expression level of tissue NOS was lower in the well differentiated than that in the moderate differentiated or in the low differentiated lung cancers. The NO level in tissues of lung was significantly higher (P<0.05) in tissues of lung cancer than that in distant tissues.Conclusions: The present data suggest that the overexpressed of NOS for stepped-up NO production in lung cancer tissues may be related to the killing of cancer cells.
