应用PCR-RFLP方法检测新生儿HIE患儿的ACE基因多态性

目的探讨新生儿缺氧缺血性脑病与血管紧张素转换酶基因多态性的相关性。方法设新生儿缺氧缺性脑病患儿为观察组,正常新生儿为对照组。分别提取外周血白细胞DNA,然后用ACE引物进行聚合酶链反应(PCR),对产物进行电泳,根据限制性片段长度多态性(RFLP)所致不同的迁移率判断基因型。结果检测出ACE基因分为DD、ID、II三种类型。观察组与对照组ACE基因型分布频率不同,观察组ACE基因型以DD型为主,与对照组比较,P<0·01,差异非常显著。结论新生儿缺氧缺血性脑病与血管紧张素转换酶基因多态性具有密切相关性,DD型基因可能为新生儿缺氧缺血性脑病的易患基因型。

Analyzing the correlation of hypoxic-ischemic encephalopathy of the newborn and polymorphism of the angiotensin converting enzyme gene employing the methods of PCR and RFLP

Objective To analyze the Correlation of Hypoxic-ischemic Encephalopathy of the Newborn and polymorphism of the Angiotensin Converting Enzyme Gene. Methods newborns with hypoxic-ischemic encephalopathy served as observing group, normal newborns served as control group. We draw DNA of lencocyte ,then we amplificate the ACE gene, using the DNA polymerase chain reaction technic. The amplification products were employed electrophoresis, at last, we analysis the genotype of the ACE according to different movement velocity caused by restriction fragment length polymorphism. Results There are three kinds of genotypes of ACE:DD, ID, II. Of the observing group, the distribution frequency of the ACE genetype DD is different from the distribution frequency of the control group. Analysised by statistacal method, DD genotype distribution frequency is higer than control group, P<0.01, the difference has statistacal significance. Conclusion We find the significant association between the ACE genotype and HIE of the newborn, DD genotype is susceptibility gene of HIE.
