芍药苷联合奥沙利铂对肠癌细胞增殖和凋亡的影响

目的：观察芍药苷联合奥沙利铂对人结肠癌细胞SW480体外增殖及凋亡的影响,初步探讨其作用机制.方法：MTT法检测芍药苷、奥沙利铂单药及联合用药对结肠癌细胞SW480增殖抑制的影响,流式细胞仪检测细胞凋亡率,Western blot检测Bcl-2蛋白表达,ELISA检测细胞核内NF-κB转录活性.结果：芍药苷及奥沙利铂呈浓度依赖性抑制SW480细胞生长,两药联合具有协同抑制作用（P＜0.05）；联合用药组细胞凋亡率为（34.2±1.43）％,明显高于芍药苷单药组（10.2±0.16）％及奥沙利铂单药组（22.3±0.22）％（P＜0.05）；Western blot法检测表明芍药苷能够显著抑制Bcl-2蛋白的表达（P＜0.05）；ELISA法检测表明,与对照组（2.013±0.04）比较,芍药苷单药组（1.17±0.17）及联合用药组（0.9±0.08）吸光值明显降低（P＜0.05）.结论：芍药苷能显著提高奥沙利铂的抗肿瘤作用,此协同作用主要是通过抑制NF-κB的转录活性,从而下调Bcl-2的蛋白表达,促进肿瘤细胞凋亡来实现的.

Effects of paeoniflorin combined with oxaliplatin on the proliferation of SW480 cells

Objective：To observe the effects of paeoniflorin combined with oxaliplatin on proliferation in human colon carcinoma cell lines SW480 in vitro and investigate the mechanism.Methods：Proliferation inhibitory rate of paeoniflorin and oxaliplatin was examined by methyl thiazolyl tetrazolium （MTT） assay,flow cytometry （FCM） was used to determine apoptosis.The expression level of Bcl-2 was examined by Western blot,the intranuclear expression of NF-κB was confirmed by ELISA assay.Results：Paeoniflorin and oxaliplatin significantly inhibited the growth of SW480 in a concentration dependent manner,and the synergistic effect of proliferation inhibition was observed when paeoniflorin combined with oxaliplatin were administered （P ＜ 0.05）.The apoptosis rate of SW480 cells treated with paeoniflorin combined with oxaliplatin was significantly higher than those treated with paeoniflorin or oxaliplatin alone （P ＜ 0.05）.The anti-apoptotic protein Bcl-2 was downregulated and the activity of NF-κB in nuclear was inhibited by Paeoniflorin,which was confirmed by Western blot and ELISA （P ＜ 0.05）.Conclusion：Paeoniflorin combined with oxaliplatin has synergistic effects in inhibiting the growth and inducing apoptosis of SW480 cells through the inhibition of NF-κB activation and the resultant decrease in the levels of Bcl-2.
