Abstract: | A series of aliphatic alcohols were tested for ability to potentiate the toxicity of carbon tetrachloride (CCl4) in mice. The alcohols were given orally at 50% of the LD50 value, 20 hr prior to an ip injection of 0.0075 mg/kg of CCl4. Using serum glutamicpyruvic transaminase (SGPT) activity to assess the degree of hepatotoxicity, methanol, ethanol, and isopropanol displayed potentiating ability and produced elevated serum enzyme activities. The most marked potentiation was produced by isopropanol. Administration of the alcohols or CCl4 alone did not result in similar changes in SGPT values. In an attempt to characterize the apparent differences in potentiating ability, liver triglyceride concentrations, liver glucose-6-phosphatase activity, and SGPT activity were measured in the rat after giving ethanol or isopropanol 3–48 hr before 0.1 ml/kg of CCl4. Isopropanol (2.5 ml/kg) was found to produce a more marked change than ethanol (5.0 ml/kg) in all 3 parameters studied. In addition, in fed animals, isopropanol plus CCl4 was singularly capable of producing hyperbilirubinemia, an effect not elicited by ethanol plus CCl4, or CCl4 alone at a 10-fold increase in dosage. |
