子宫内膜癌细胞凋亡与p53蛋白异常和细胞增殖的相关性研究

摘    要:目的 探讨子宫内膜癌组织中细胞凋亡与p53蛋白表达状况和细胞核增殖的影响,与子宫内膜癌组织学类型和恶性程度的关系。方法 采用DNA末端转移酶介导的Bio-dUTP原位末端标记技术和免疫组化技术及HE染色,对细胞凋亡与p53蛋白表达和增殖细胞核抗原(PCNA)进行原位和相关性研究。结果 64例子宫内膜癌组织中凋亡细胞密度均低于对照组,并随肿瘤恶性程度及p53蛋白表达升高而降低。64例子宫内莫膜癌46.9%表达p53蛋白,阳性细胞含量随肿瘤恶性程度升高而增加,对照组全部阴性。结论 提示子宫内膜癌p53蛋白表达和功能异常与其细胞增殖及凋亡失衡有关。

Abstract:Objective:To explore the effect of p53 protein expression in endometrial carcinoma on the cell proliferation and apoptosis,and the relationship between above parameters and histological classification and malignant degree of this carcinoma.Methods Using terminal deoxynucleotidy1 transferase-mediated dUTP nick end labeling(TUNEL)technique,protein p53 expression,proliferating cell nuclear antigen(PCNA)immunohistochemical staining,and HE staining,we observed and compared the index number and distrbution of apoptotic cell,protein p53,proliferating cell in situ in 10 cases of normal endomertrial tissue and 64 cases of endometrial carcinoma tissue.Meanwhile,we also made a correlation studies.Results (53 13%)of the endometrial carcinomas expressed p53 protein.The amount of carcinoma cells positively stained for p53 protein increased with the degree of malignancy.All the control group did not express p53 protein.The densities of positivity for proliferation cell nuclear antigen staining in endometrial carcinoma group were higher than that in control group.On the contrary,the density of apoptotic cell in endometrial carcinoma group was lower than in control group.Conclusions These results suggest that above 3 parameters have a referential value for the evaluation of bilogical behavior of endometrial carcionmas.It is clear from the present study that the abnormality of expression and function of p53 protein correlates with the imbalance between cell proliferation and apoptosis.This may be one of the important factors in the development and progression of endometrial carcinoma.
