响应面法优化车前子油的提取工艺及抗氧化活性研究

摘    要:目的:优化超临界CO2萃取车前子油的最佳工艺条件,研究车前子油中脂肪酸的组成成分和含量以及其抗氧化活性.方法:在单因素实验基础上,利用响应面法,以萃取率为响应值,建立数学模型,获得最佳工艺;利用GC-MS分析车前子油中脂肪酸组成及含量,并采用自由基清除能力体系初步评价车前子油的抗氧化活性.结果:通过二次回归模型响应面分析得出最佳超临界CO2萃取车前子油最佳工艺条件是萃取温度70℃,萃取压力30 MPa,萃取时间120 min,CO2流速30 L·h-1,在此最佳工艺条件下车前子油萃取率为35.07%,理论值为35.91%,与理论值相对误差小于5%.经GC-MS分析得车前子油主要成分为亚麻酸、亚油酸、硬脂酸和棕榈酸,其中不饱和脂肪酸含量超过88%.抗氧化活性研究中,由DPPH+和ABTS+自由基清除实验得IC50分别为1.13,3.57g·L-1.结论:由响应面法优化得到的车前子的提取工艺方便可行,得到的车前子油具有较强的抗氧化活性,值得进一步开发应用.
