人胃癌及胃间质细胞肿瘤Epstein-Barr病毒的检测

目的 探讨Epstein-Barr病毒感染对人类胃肿瘤发生，分化程度及淋巴结转移的影响。方法 取冰冻组织及石蜡切片提取DNA，用PCR方法扩增EBV-DNABamHI-W片段，阳性者提示有EBV感染，分析EBV阳性胃肿瘤的分化程度，淋巴细胞浸润和淋巴转移情况。结果 109例胃癌中共有18例检出EBV-DNA（阳性率为16.5%)，3例胃间质细胞肿瘤中有1例EBV-DNA阳性（阳性率为33.3%)。EBV阳性胃肿瘤分化程度低，淋巴结转移率低（r=-0.176,P=0.031)。富于淋巴细胞浸润，结论 EB病毒感染可能参与胃肿瘤的发生。Epstein-Barr病毒感染与淋巴结转移率呈负相关。

Detection of Epstein-Barr Virus in Human Gastric Carcinomas and Gastric Stromal Tumours

Objective: To investigate the influence of Epstien-Barr virus infection on carcinogenesis, differentiation and lyphaden metastasis of gastric neoplasm .Methods: We extracted DNA genomes from frozen tissues and paraffin sections.Polymerase chain reaction(PCR) was used to amplify 129-bp fragment in BamHI-w region of EBV-DNA and the presence of EBV-DNA was evaluated.We analyzed the condition of differentiation, the infiltration of lymphocyte and lymphaden metastasis of EBV-positive gastric neoplasm. Results: EBV was observed in 18(16.5%) of 109 gastric carcinomas and in 1(33.3%) of 3 gastric stromal tumours .The rate of lymphaden metastasis in EBV-positive gastric carcinomas was lower than that in EBV-negative gastric carcinomas( r=-0.176, P=0.031).The EBV-positive gastric carcinomas was liable to be infiltrated by lymphocyte. Conclusions: EBV infection possibly affected the process of gastric carcinogenesis and negatively correlated with lymphaden metastasis of gastric carcinomas.
