摘    要:目的建立尽量接近人类自然条件下发生的急性病毒性肝炎感染动物模型,研究其发病机制和治疗方法。方法选择中国地区高流行的人HBV C基因型具有复制能力的1.2倍HBV基因组为材料,构建可高效表达HBV基因组的重组腺病毒相关病毒载体pZAC2.1-HBV。利用尾静脉水动力注射法将pZAC2.1-HBV注入小鼠。结果瞬间转染人肝癌细胞系HepG2细胞后,在细胞上清和裂解液中检测到HBsAg。ELISA方法检测小鼠血清中HBsAg和HBeAg的水平可持续表达14天,随后抗-HBs、抗-HBe出现。同时在14天之内可检测到血清中HBV-DNA特异片段。结论采用水动力注射法成功建立HBV感染急性小鼠模型。

Abstract:Objective To set up a mouse model infected by human HBV C genotype DNA,and to investigate its mechanism and potential therapeutic strategy. Methods A 1.2-genome length HBV C genotype inserted in AVV vector,the recombinant expressed vector pZAC2.1-HBV was successfully constructed. Results After transient transfection in HepG2 cells,HBsAg was detected in cell supernate and cell lyses.By hydrodynamic injection of HBV-DNA into the tail veins of C57BL/6 mice,HBsAg and HBeAg were detected in serum of C57BL/6 mice up to 14 days,meanwhile HBV-DNA specific fragment was detected in serum up to 14 days. Conclusion HBV transfected mice model is successfully set up by hydrodynamic injection of HBV-DNA into the mice.
